CCM: CAS reveals new mechanism of plant hormone signaling pathway 09-13-2016

The CAS reveals new mechanism of plant hormone signaling pathway by successfully analyzed the chelation structure of strigolactone metabolite and DWARF14 active site, the crystal structure of complex of receptor DWARF14 and hormone GR24, and discovered the destabilization of DWARF144 and homologous protein DAD2 in the existence of hormone.

A research team led by Xu Huaqiang from the Shanghai Institute of Materia Medica, Chinese Academy of Sciences (CAS) jointly with a research team led by Li Jiayang from the Institute of Genetics and Developmental Biology, CAS, etc., successfully analyzed the chelation structure of strigolactone metabolite and DWARF14 active site, the crystal structure of complex of receptor DWARF14 and hormone GR24, and discovered the destabilization of DWARF144 and homologous protein DAD2 in the existence of hormone, revealing a new mechanism of strigolactone signaling pathway.

The related achievement was published online in Cell Research titled Destabilization of strigolactone receptor DWARF14 by binding of ligand and E3-ligase signaling effector DWARF3.

Strigolactone, an important plant hormone discovered in recent years, has important effects of awareness and signal transduction during the process of plant growth and development and environmental adaptation. There is still something ambiguous like the structure, function and the mechanism of signaling pathway of strigolactone receptor. So researches of genetics, biochemistry and structural biology of strigolactone will provide structural basis for developing new herbicides.

The newest achievement is a significant breakthrough in the research field of strigolactone molecular mechanism contributed by research teams led by Xu Huaqiang and Li Jiayang. They have published three academic papers about this field in 5 years. The first paper was published in Cell Research and the relevant achievement analyzed the 3D structure of strigolactone receptor protein D14 and homologous protein KA12 which provided structural basis for further elucidation of strigolactone molecular mechanism. And the second paper was published in Nature which elucidated that strigolactone can regulate rice tiller and plant type by inducing the degradation of its inhibitor D53.
